| A position is open for a PhD student or for a pro gradu student that is motivated to carry out post-graduate studies. The purpose of the project is to develop new methods for enrichment of adult stem cells for clinical purposes. Stem cell culturing, antigen/antibody production and developing techniques for antibody utilization are examples of the methodologies that will be used during this project. The research project is a joint of three different research groups of the University of Oulu: Professor Petri Lehenkari (Anatomy); Professor Olli Vuolteenaho (Physiology); and Professor Juha Risteli (Clinical chemistry). The project includes close collaboration also with other research units and companies in Finland and it is funded by TEKES SHOK network (SHOK = The Strategic Centres for Science, Technology and Innovation). The general aim of the project is to advance current knowledge of the structure-function relationship of adenosine receptors and transporters, and their signalling pathways in the nervous system (http://www.ncbi.nlm.nih.gov/pubmed/20538452). We are looking for a motivated student who is approaching the pro gradu phase of his/her studies. The applicant should have some experience in basic recombinant DNA techniques, protein analyses and cell culture. The project will mainly include “driving-in” Leishmania tarentolae expression system in our laboratory (http://www.jenabioscience.com/cms/en/1/browse/1007_eukaryotic_expression_system_ lexsy.html). The project can be started in late summer of 2010 or later in the autumn. Details are negotiable. | No mutta hei! Fuksisuppi on nyt julkaistu netissä, valitettavasti painettuja fuksisuppeja on vain yksi kiltahuoneen arkistoissa. Suppeja tulostettiin yhteensä 60 lähetettäväksi koulutusohjelmaan hyväksytyille. | Pro Gradu project in Bioprocess engineering laboratory (University of Oulu) A Pro Gradu project is available for a student of Biochemistry with a title: Directed Evolution of Triosphosphate Isomerase (TIM) towards New Substrate Activity Background The project is part of a BioCat joint project (including groups from the University of Oulu and the TU Berlin, Germany) which aims at the development/creation of tailor-made engineered biocatalysts to be used for the efficient, environmentally sound, biotransformation of chemical building blocks into highly pure enantiomers. On the basis of a known framework (the TIM barrel protein Triosephosphate Isomerase) an artificial biocatalysator will be designed. While keeping the catalytic activity the substrate spectrum is changed. This project aims to convey the catalytic competence of a D-xylose isomerase and L-arabinose isomerase on an engineered monomeric TIM variant called A-TIM. Techniques and experimental approach The methodology for this project will be mainly molecular biological: different kinds of PCR, sequencing, transformation, cloning, expression and cultivation optimization etc. The aim is to create libraries of A-TIM through a random approach via epPCR and a targeted approach using a megaprimer PCR method. The plasmid vector to carry the libraries will be optimized for the screening and the expression. To screen the libraries a suitable knockout strain will be created based on the protocols by Datsenko and Wanner (2000) and Thomason et al. 2007. The libraries are screened in vivo on agar plates with xylose or arabinose as the only carbon source. The student will have the chance to familiarize him-/herself with directed evolution methods which are state of the art techniques in protein engineering. | There are projects available for 1-2 Pro gradu students in the group of Inari Kursula at the Department of Biochemistry, University of Oulu. We are looking for highly motivated candidates with a keen interest in protein chemistry. Experience in both protein purification and cell culture methods is a definite plus. The successful candidate can work also independently and is not afraid of challenging projects. Our research is focused on actin-binding proteins of the malaria parasite. We use X-ray crystallography and small-angle scattering of X-rays for structure determination and biochemical and biophysical methods, such as light scattering, SPR, ITC, fluorescence and CD spectroscopy, for the characterization of proteins and protein complexes. The available Pro gradu projects would involve constructing co-expression systems for parasite actin and actin-binding proteins, mutagenesis studies, and optimization of protein expression and purification conditions in both bacterial and insect cell systems.
